First of all use standard phytochemical methods to detect their presence (Look for this resource: Trease and Evans) then continue with GC-MS where the spectral properties of the phytocompounds can be compared to standards to help you determine the functional groups and the possible chemical structures.
GC-MS and LC-MS (correlation with literature spectra, RI values etc. ) can be used for tentative identification of a compound, but if you really want to be 100% percent sure in its identity you need to either:
1) Isolate the compound and elucidate its structure using NMR (1D and 2D) and X-ray (if possible).
2) Synthesize it, confirm the structure of the prepared compound by NMR and then prove that the compound is present in the plant extract by co-injection.
3) Buy the compound and do the co-injection of the standard with the extract like in 2).
