There are problems germinating old transgenic rice seeds (Japonica). Some seeds are 4 years old, while others are 3. They were stored at room temperature. Can we treat them in a way to have at least 20-30% germinate? Please let me know.
I think the first thing you should do is to select within those rice samples the best kernels for germination. There is a very easy and effective method proposed by Japan International Cooperation Agency, which I add in the links below:

Go for seed priming @15 0C, for 24 hours. This will lead to repair of damage due to seed ageing. You can expect 30-40 % higher germination compared to control. Alternatively you can go for tetrazolium test to accertain if the seeds are not dead.
