There's a human membrane receptor X, for which we do not know the natural ligand.
We know a human membrane receptor R which exhibits a high homology with X at the level of the primary sequence.
We also have a ligand for receptor R, i.e. protein RR which does not bind to X.
I want to find the ligand for X.

You might try do a pull-down experiment to find candidate proteins that bind to Xr. If the sequence of Xr indicates that there is a large continuous sequence constituting an extracellular domain, then it would be worthwhile to try to express this domain as a soluble protein, which would be easier than doing the experiment with the whole membrane protein.
