I have an image obtained though Particle Image Velocimetry (PIV) which contains both particle and fluid. Is there any tool or process available in Matlab or Python that can give me the volume fraction of particle in that particular image?

You can simply use the particle analysis tool in ImageJ (an open source software). But please be aware that the volume should also consider the light sheet thickness.
OpenPIV is an open source Python software (disclaimer - I am one of the developers) and it's pretty easy to extend to get what you're looking for. Indrajit Nandi

we use scikit-image for the image processing and it's a very rich set of tools
I suggest to infer the dimension from the acceleration of the particle deduced from PIV measurements and an hypothesis of the geometry (sphere ?).

You can either use Image or Matlab.
- First thing is to binarize your image such that particles are white (255) and the background is black(0). For the correct selection, you may have to adjust the threshold. I suggest you try in ImageJ, its much easier there.
- Then check the number of white pixels (intensity = 255, resembling particles) and black pixels (intensity =0 resembling background).
- Calculate area fraction = # of white pixel/(# of white pixel + # of black pixel).
Remark: Note that this is 2d volume fraction.

You can use the Trackpy feature detection method (written in python).
Or by using Fiji > analyze > analyze particles tool.
As Hisay mentioned above, whatever you calculate from a single image will be the area fraction.
