First homogenize some digestate in blender. Then dry a certain amount of digestate at 105 degrees centigrade. Pour a certain amount of dry matter into the COD tube. Start the experiment. Find the COD value of 1 gr from the resultant ratio.

Take a composite sample for at least 24 hours. You can take an small amount each hour or 30 minutes.

Then homogenize and analyze.

The result will be different if you grind or not the sample, depends on what you want.

In addition, I would like to point out that in order to reach the most accurate result, you have to continue with the experiments until the standard deviation of COD results is the lowest (among the homogenize samples).
