One of my friend is trying to extract WBCs and stem cells from the whole cord blood using lysis buffer. He wants to remove RBC and hemoglobin. Can anyone help him with the protocol or any suggested method he can use to do the same?

last time i use Magnetic Activated Cell Sorting (MACS) for stem cell separation , but for both maybe u can combine with conventional method

You could perform a purification using ficoll gradient. It will seperate mononuclear cells from the other cells (platelet and RBC). You will have less RBC and, after, increase the proportion of stem cell (which one ?) by magnetic purification.
Alternatively, you could use RosetteSep to enrich your sample in stem cell by elimination of unwanted cells.
