You should emulsify the protein or peptide of interest with complete Freund's adjuvant (repeat 1 additional time) and a dose of the protein of interest and incomplete Freund's adjuvant for a given period of time, include mouse controls without this treatment but applying a vehicle intraperitoneally (Physiological saline solution), then titrate both IgM and IgG by a home ELISA, sensitizing the wells with the protein of interest.
Remember to include as a control the protein alone in the buffer you select.
