Directly can be added 1 mg/ml in PBS. If you want higher concentration, take required amount of ANS and add a drop of methanol to solubilize and make up the volume with PBS.
ANS is soluble in organic solvents such as DMF and DMSO. However, to dissolve ANS in PBS you can achieve that in either of the following ways bear in mind that each has pros and cons:
1. To obtain an organic-solvent free aqueous solution of ANS, dissolve it directly in PBS. Note that the solubility of ANS in PBS pH 7.2 is ~ 1mg/mL.
2. Alternatively, the solubility can be improved (up to 20 or 30 mg/mL) by dissolving the ANS in either of the aforementioned organic solvents first, then you can prepare the desired concentration in the aqueous buffer from the stock by dilution.
It is important to purge the organic solvent with argon or nitrogen prior to the preparation of the stock in the organic solvent.
