I'm trying to dissolve hexane extracts of some plants. The extracts are very viscous, and I need make solutions at 50mg/mL. After, I will test them in in vitro biological assays. So, the solvent mustn't be too agressive.
It already have been tested: Ethanol 90%, Methanol 75%, Acetone 70%, Tween 80 (10%) and DMSO (100%). None of all dissolved the extracts completely.
Can anyone suggest me other solvents?
I think hexane extract contains some fats and waxes , so first remove the fats from the extract then try again to dissolve .
There is no solvent can be make the extraction completely . This due to the multi components of molecules in each plant extraction.you can use pure pentane as non polar solvent and dioxane in different ratio.
The extract was originally dissolved in a non-polar solvent (hexane) and many of those compounds are only slightly soluble in the polar solvents you described. You can pick up the sample in your solvents that work with your assay and filter them to remove the solids; run hexane through the filter to capture the solids afterwards if desired.

For such extracts works for me well in hot DMSO and using ultrasound
You can try dissolve the extract in hexane, and then mix with other solvent that is suitable for biological system.
You could try to dissolve the extract in mineral oil. Thereafter dissolve the solution again as the in vitro assay recommended.
these extracts must be dissolved in the liquid CO2 to the desired kollichestvom solvent under conditions to critical and super-critical conditions for CO2. Then CO2 is removed - and the solution remains.
