I have used several bacterial strains for removal of heavy metals from a contaminated source. After measuring the initial and final concentration of heavy metals there is a significant difference observed. Now i want to develop adsorption isotherm using these data considering different environmental conditions e.g. pH, temp etc. How can i develop a adsorption isotherm using these data and fit them into Langmuir isotherm model and Freundlich isotherm model?

First, you need to the optimized maximum adsorption capacity of your adsorbents for adsorbate (heavy metal) by varying initial adsorbate concentration from 0 to until you achieve equilibrium. Then you will get a data set and you can apply Isotherm on that data set. you can use different software for isotherm modeling such as MATLAB, Origin or even Excel.
if you consider bacteria as adsorbents, you can do as described above:
1) Determine the maximum sorption capacity.
2) Carry out a series of experiments at various concentrations of heavy metals in the range 0 - the concentration corresponding to the maximum adsorption capacity.
3) Get a few isotherms for different parameters of the experiment.
