I have some XDR and PDR bacteria, when i test ESBL production by combined disc method, the bacteria was resistance to cefotaxime and also resistant to combined disc cefotaxime/clavulanic acid, no inhibition zone for two discs. How i can test ESBL for these isolates?
PCR-based detection of ESBL genes (e.g., bla_CTX-M, bla_TEM, bla_SHV) is the most reliable way.

Depends on what exactly you want to test for and what are your available methods as well as organisms of interest.
I assume its Gram-. If you want to know in a targeted manner what are the lactamases present, PCR or whole genome seq is the best approach.
If you want to use a phenotypic testings, and you have MDR organisms with resistance to clavulanate and CXT or FOX, then it is slightly more sticky. This tells me that you have most likely some carbapenemases present, therefore you have to use a EDTA, phenylboronic acid, avibactam and aztreonam as a screening strategies.
