We are currently producing BMP-2 and we would like to determine its ED50 through an in vitro assay, if it is possible. As far as, I am concerned, it should be done through an alkaline phosphatase dose-response assay. However, I am not sure, which is the statistical power we need to do it, so as if there is any consideration we need to have into account.

ED50 or EC50? , i.e. it is ofr in vivo (ED50) or for in vitro (EC50) assay

Then you need a protocol of measuring , for instance, the level of expression (if a mRNA) or the amount of BMP-2 (by Western perhaps?) at different concentrations of the "inducer" (the compound for which you want to calculate the EC50). Then you plot the amount of mRNA or amount of BMP-2 (Y axis) versus the concentration of the "inducer" (X axis). Then there are programs, even on line in which you put the Y and X values and you get the EC50.

If BMP-2 is an enzyme and you want EC50 for an activator the approach would be different. But I assume that you are nt working with any enzyme.

BMP-2 is a morphogenetic protein, produced in our lab.
We have made some experiments and determined EC50 as you told me, same as it is stated in bibliography.
