I want to know how much beta-carotene are there in each 100 grams of tomato, spinach, and cantaloupe. If I obtain 100g of each fruit/vegetables, take 10g sub-sample from each, extract their beta-carotene and test it using spectrophotometer. How would i prepare a standard solution and calibration curve for it to know its concentration in microgram/g? Let's say I add 10mg of beta-carotene in 100 mL kerosene. (also can i use kerosene as an alternative for hexane?).

I think hexan and petrolum ether are best solvents for carotenoids.
When prepare serial dilution for beta carotene you can start from the solution you choose then prepare from it a serial dilution with little concentrations you choose (little than 10 mg/100ml) for example choose (1- 2 - 3 -4 mg /100 ml) then measure their absorbances to make the curve of serial dilution.
You can see how another studies prepare this serial dilution and start with the concentrations they depend on.
when you have the equation of serial solution, the concentration you get in mg/100 ml is the same concentration of carotene in 10 g of sub sample. then you convert it to ug/g easily.

so you can use vegetable oils like olive oil for a green extraction by ultrasonic or by a microwave.I did not find any study for using kerosene in extraction.
