I want to modify disposable carbon and gold SPE's with gold nanoparticles to prepare an immunosensor.
What is the easiest method to achieve this?
Do I need a pre-activation step for electrodes to obtain a better signal?
If I dip my SPE completely into a solution containing Au nanoparticles and apply a potential for Au-np deposition, doesn't it effect reference and counter electrode? What should I do to protect these electrodes?
If these electrodes are disposable, can I test CV curves after preactivation, nanoparticle modification and antibody immobilization and antigen detection peaks with the same electrode?
Could you please inform me about any of these?

As you rightly suspect, it is always a good practice to start with a cleaned electrode surface prior to any surface modification or measurements. For SPE, this can be easily obtained by electrochemical ’activation’ or cleaning in sulfuric acid (by applying anodic potentials) to remove layers due to high polymer in the manufacturing ink or impurities resulting from poor handling. This will ensure an optimal electrochemical performance of the SPE.
As regard dipping of the electrode in solution, I believe there should be no negative effect on the reference and counter electrode, especially if the solution is not acidic. However, if you deal with harsh solution you can be sure the reference electrode will be affected. In this case I will suggest you try other AuNP immobilization techniques such as the use of self-assembled monolayer using thiol moiety for the SPE-Au.
Yes, you can very easily follow your electrode modification steps on the same electrode. The only challenge may be in recovering and/or re-using the same electrode after all of these modifications. I hope these help.
