I would like to set up a colony formation assay and do a combinational drug treatment on a WT and Knockdown variant of a lung cancer cell line. For both cell lines, I have done the biological replicates of each individual drug and calculated the average IC50s using the CompuSyn software. My question is how do I determine the dosing? Should I use a fixed ratio or non-constant ratio? One of the drugs is Cisplatin and my interest is monitoring the change in Cisplatin sensitivity in the KD compared to the WT when the 2nd drug is added. Any insight to this would be appreciated!
