Dear experienced researchers,
I have received the comments from editor and reviewers for my revision. The editorial decision was minor revision once again as "One of the reviewers has raised questions that need to be addressed in a revised version, for the manuscript to be taken any further. Please submit a revised version addressing the concerns of this reviewer". But Re-read comments of this reviewer several times, they are not difficult to answer but scientifically out of scope of our study. I have already politely explain this in first revision.
So I wrote to assistant editor, he briefly answered me that he can't do anything and must follow the instructions !!
I'm confused what I do ? email the editor-in-chief or withdraw the manuscript ? I would be very grateful if the experts help me
Editors and reviewers are not infallible; on the other hand, there should usually be a reason why the remark has been made.You may think it irrelevant, but the reviewer obviously did not think so. Put yourself in the reviewers' shoes and wonder why he made the remark in the first place (often it is from a personal experience).
If you find the comment really irrelevant, then the best solution is to answer the reviewer (e.g. thank you for the remark, but in the framework of the article it seems not so relevant). Alternatively, you ask the handling editor for en explanation. It is your article, your study, and ultimately you are responsible for it.
In the biomedical world it is usually not a good idea to withdraw a manuscript if it is under consideration for publication in a reputable journal. It would delay publication significantly.
Immediately withdraw the MS and submit elsewhere. A good Editor will always explain the author with logic.
If you answered the reviewer's comments with supporting scientific arguments based on literature citations and still is not acceptable by both the reviewer and editor, submit it to another journal.
Ask to be referred to another reviewer, if possible! If not, just withdraw your manuscript and submit it somewhere else. Good luck.
This may happen if the reviewer is far from your profession and editors may accept only the reviewer suggestion.I have experience in this regard and the editor respond to me by saying "I simply accept the reviewer comment not to reject them". Therefore, if you faced this kind of response go to another journal.

In my view, if an editor accept the reviewer rather than the actual researcher the best option would be your decision
i agree with those who suggest another journal. reviewers are consultants. a GOOD editor will, especially after a thoughtful reply by author why there is a problem, tell her reviewers, "thanks for sharing" and do what she - like managers, supervisors, directors, conductors et al. - is supposed to do and make an informed executive decision. if she can not explain in her own words why the author needs to bend to suggested revisions, i would withdraw paper. i have done this many times as recently as a few months ago. and i always tell the editor the logic behind my withdrawal.
