What is an easy method to count total leukocytes in rat blood? Can I just use a hemocytometer? Does anyone have any good images/protocols for what to look for? Do I need any special equipment? I am not familiar with this type of technique but want to compare relative leukocyte counts between two treatment groups to support evidence of leukopenia.

METHOD:-
Hematologic analysis
WBC, PLT, and RBC counts were obtained from the EDTA-anticoagulated whole blood samples using an ADVIA 120 analyzer and Multispecies Software (Ver- sion 3.1, Siemens Medical Solutions, Norwood, MA, USA). Blood smears were stained with modified Wright–Giemsa (Advia S60 Auto Slide Stainer, Sie- mens Medical Solutions). A Nikon Eclipse E400 micro- scope (Nikon Instruments Inc., Melville, NY, USA), with a 22-mm field diameter, was used to estimate cell counts. Cells were counted in 10 fields within the monolayer of each smear and the mean number of cells per field was calculated. All blood cell estimates were performed by the same laboratory investigator (CSD). Leukocytes were counted using a 20 objec- tive in 60 smears from 19 studies, PLTs were counted using a 100 objective in 58 smears from 18 studies, and RBCs were counted using a 100 objective in 65 smears from 15 studies
