I am extracting bioactive components from Aloe vera gel and its powders.
I am after some calculations to confirm
In the case of gel I took 10 g of whole leaf Aloe vera gel (liquid form ) in 10 ml methanol, and in the case of powder tool 1g of powder in 10 ml methanol, done extraction by an ultrasonic bath (but haven't evaporate methanol), straight away took 2ml filter and did HPLC. suppose by standards curve I got x=0.146mg/ml
how I can do back calculations to quantify it in mg/%TS, to compare both as one is liquid and the other is a powder form of the sample?
You have estimated the conc. to be 0.146 mg/mL. Total volume for extraction is 10 mL. That means you have 1.46 mg in 10 mL. This has come from 10 g material (doesn't matter if it was a gel or powder). So, 10 g material has 1.46 mg, then 100 g will have 14.6 mg, or 0.0146 g. The content is 0.0146%.

It may not be possible to get good results from the HPLC analysis without evaporating the methanol. Therefore, after filtration, you can easily evaporate the methanol with the help of heat and dilute the remaining residue with the mobile phase before applying it to HPLC.
