I am trying to express a recombinant protein in the yeast pichia pastoris in a laboratory bioreactor scale under fed batch condition. i want to know how to set the external peristaltic pump ( non related to the bioreactor core) to do an exponential feeding rate. the feeding rate is calculated using this equation:
F(t)SF= ((µ / YX/S)+ms)v0 xt eµ(t-t0)
The answer depends on the type of pump and whether it is able to accept a programmed feeding rate. Typically, peristaltic pumps have only a rudimentary feeding scale (from say 0 to 10), which you need to calibrate off-line and then adjust as the fermentation proceeds. The other way, which is more accurate, is to place the feed (in its feeding container) on a digital scale and then use the output from the scale to switch the pump on and off, based on your algorithm. The latter requires that you can connect the scale to a PLC or a computer, and then switch the pump based on the changes in mass.
