To elaborate on the question:
1.Why don't some prospective supervisors reply to emails even when they have advertised some open PhD positions on their website?
2.After sending in the first inquiry,how long should one wait before sending a follow up email and what information is necessary in this follow up email.Should one resend the entire first email or simply ask if the first email was received?
3.Any other necessary advice on how to get the response is highly appreciated.

Maybe they are busy, maybe do not find your background or expertise appropriate for that position, maybe the position has been filled already but the web site has not been updated yet.
But give them a call, sometimes they respond and it works better than email.
