Does anyone know a cultivation technique, where you pour agar media over liquid media? I was told to do so as it the best way to do next experiments. At least this is what I understood. Or do I need to pour just agar in falcon and then transfer some bacteria with loop while it is liquid?
So I am cultivating Lactobacillus acidophilus and using MRS media. I will use falcon tube.

it is not possible to mix broth media and agar media in one flask at a time. however if you are separately use for lactobacillus, the best way for soldifiying bases with agar media in glass test tubes for isolation and culturing and maintenence of lactobacillus with optimum conditions. based on that your streaking skills you can use your required glassware.
