Recently, I plan to detect some mRNAs expressed level of whole life stage of maize weevil, but I don't know how to collect it's eggs. So does anyone have some experence of collect the eggs of S. zeamais, S. oryzae or S. granarius ?
Yes I have collected Sitophilus eggs. They are always laid inside grain so I chipped away the grain from around them using a needle to leave them standing proud. It takes patience. Take a look at these papers on my RG site:
(PDF) Egg Size and Reproductive Strategies in Insects Infesting Stored-Products (researchgate.net)
You can locate the egg plugs underneath which the eggs sit by staining:
(PDF) The effect of increased grain moisture content on some life history characters of Sitophilus oryzae (L.) after staining egg plugs with acid fuchsin (researchgate.net)
The eggs can most easily be extracted when laid close to the soft germ in wheat. If you can't locate the papers just let me know. Good luck.
