Currently, I would like to study more about seed development and apomixis. I want to check the ploidy of endosperm of seed in the case apomixis happens. I know one way is to determine by using flow cytometry. But my lab is not keen on cytology. I wonder if there is another simple way or not? Thank you so much for reading my question.

Actually there is no simple way to determine ploidy for sure. Other methods would be linkage analysis in a segregating population or counting the chromosomes. Flow cytometry is the most simple. You just need a (if available haploid), diploid and polyploid control to compare your tissue with these controls.
the best way is compare the diploid form with the polyploidy form. but for identifying ploidy beside flowcytometry you can study anatomy of the plant which you want to determine its ploidy and compare with progenitor.
