I am doing imaging experiments on samples immersed in 400uL media in an 8-well dish. The goal is to image z-stacks of multiple positions through time (xyzt, mark and scan). We have a Leica DMI6000 with the Leica Application Suite X installed.

I have an issue where the z-stack changes over time, so gradually my samples in each position aren't imaged in the correct z-dimension range. We have tried using autofocus, but the fluctuations may be too small for it to work?

We would like to slow down the speed of the stage as it changes positions. We think the z-dimension fluctuations are arising from too rapid stage movement that causes the liquid in the wells to affect the z-dimension. Does anyone know how to adjust this setting?
