I am trying to measure a biologic's effect on proliferation of a cell line. The cells secrete a stimulant to induce proliferation so I don't need to add the stimulant as a separate treatment.

I have blank wells (no cells), negative control wells (cells without the inhibitory reagent), and I want to measure proliferation after 5-7 days so I also have cells in a different plate to be measured on Day 0 to establish the baseline for proliferation.

How should I calculate % cell viability?

I can use this formula:

percentage of cell viability = (Atreatment − Ablank)/(Acontrol − Ablank) × 100%

But is there a way I should add my Day 0 data to the calculation?
