I have used 3 types of media for growing an yeast.
1. LGI + 0.005% Yeast Extract
2. LGI + 0.005% Yeast Extract + 0.5% ammonium sulphate
3. YPD (1% Yeast Extract + 2% Peptone)
Now, how do I calculate the percentage (%) or concentration (mM) of nitrogen used in respective medium?
As we know,
nitrogen in Yeast Extract - 10.5 %
nitrogen in ammonium sulphate - 21 %
nitrogen in peptone - 15.5 %
Please use S1V1=S2V2 formula for calculation. S1=concentration of pure sample (known); V1= volume of pure sample to be taken for your desired concentration (you need to know); S2=Concentration you need (known, like 1 ppm, % or mM etc.; working concentration); V2= volume you need (you have to decide; working volume).
Hi there, If YE is 10.5% in nitrogen then your first media is 0.005 x 0.105 = 0.000525% in nitrogen (which is 5.25mg/L). If AS is 21% in nitrogen then your second media will be 0.000525% + (0.5 x 0.21) = 0.105525% in nitrogen (1.05525g/L). Media 3 will be (1% x 0.105) + (2% x 0.155) = 0.415% in nitrogen (4.15g/L). Nitrogen being 14g / mole, media 1 will be 0.00525 / 14 = 0.375mM, media 2 will be 1.05525 / 14 = 75,375mM and media3 4.15 / 14 = 296.4mM.
