here i dont know which value should i consider for AbsControl
because after dilution absorbance should b 0.700 but know values of my extract is 0.2727... if i consider 0.700 than all my values are in negative form
Hi,
Abs Control is the absorbance of your negative control, so mix of everything without your tested substance. Abs Sample is your reaction mix with your tested substance.
AbsControl is the absorbance of all reagents except the extract (about 0.7), whereas absSample is the absorbance of all reagents including the extract (0.2727 in your example). You can use the equation you cited with these values and you will not obtain negative results.
