I have data for antioxidant activity on Excel sheet and I draw scatter chart with the equation: y = 1.3534 x + 0.58, and R² = 0.9927. I want to calculate the IC50 for inhibition %. Someone can help me to figure out how to calculate IC50 value?
In addition, some of my colleagues use a function called “Growth” in Excel to calculate IC50 by just by selection inhibition data column and then choose Growth from Insert Function (fx = ) and obtaining the result. Is it a correct way?
After you got the regression equation y = 1.3534 x + 0.58
you can calculate the IC50 as follows:
IC50 = (0.5 - b)/a.
IC50 = (0.5-0.58)/1.3534
You replace (y) by 50 in the equation y = 1.3534 x + 0.58 and you deduce ( x) .

Dr. Omer Ibrahim, you mean IC50 = (0.5 - a)/b, not (0.5 - b)/a? Accordingly, the result will be in minus, right. Is it the IC50 values be a minus.
firstly you more than six concentration to draw you chart and best ways to calculate it via special program-me called graph pad program-me version 6 and you can neutralize also by log factors hopefully to benefit for these programme

IC50 = (0.5-0.58)/1.3534

From your regression equation,equate to 50 and solve for x
50 = 1.3534 x + 0.58
X = 50-0.58/ 1.3534
X= 36.6
Therefore IC50 is 36.6 in this case which means 36.6 microgram or milligram (what you have taken) is the dose or concentration that scavenges half the DPPH
