There is two method for that, Mooney‐Rivlin equation and the Flory‐Rehner theory

Given the average molecular weight of an unbonded polymer and its specific volume, you can estimate the crosslink density.
First you need to measure the elastic modulus of the hydrogel - E.
Then calculate the crosslink density:
N (mole nodes per volume) = E / (3RT)
This method is based on Flory-Rehner and / or Mooney-Rivlin theory and equations.

The question is crosslinking degree of determined amide group?!
So, I would use titration of the gel by Ninhydrin. You may see the international standard ISO-9702 protocol, to determine the number of primary, secondary, and tertiary amine groups in a polymer.
For the total gel content (a measure of crosslinking) simply use Soxhlet extraction of the hydrogel.
