it is easy to calculate initial COD for synthetic wastewater having glucose or acetate as carbon source. But how will i calculate final COD which could be effected from the sludge in the same chamber.? and how will i take sample from anaerobic chamber having 500 settled sludge and 1500 ml synthetic wastewater so that it could be compare with initial.? please refer to the figure attached.

I think it could be done by taking sample from supernatant but its better to filter the sample by a specific pore size for example 0.4 micron filter and analyze the filtrate for COD. It seems to give more accurate results.

Better you centrifuge/ vortex the sample to make it more homogenous and filter through the definate pore size and use the filtrate for analysis. It is recommended to do triplicate of the test that helps to minimize the errors.

COD analysis for synthetic wastewater is easy but the chemical procedure need to avoid solid particles. This is not a problem if you create your solutions. But if you use directly synthetic wastewater with an high sludge content a solid TOC analysis is better. Generally some drop of your sample are placed in a fiberglass and than in the oven. But you need the instrument...if you don't have this possibility centrifuge and filter always, at the beginning and at the end of your test.
