How to calculate 1) Amount of drug released and 2) Cumulative percentage release (%). The amount of drug conjugated particles in dialysis bag is 5 ml, Bath volume is 150 ml and sample withdrawn is 3 ml. The amount of drug used for synthesis is 1.8 mg for 10 ml solution, out of which 5 ml solution is taken for drug release study.

Since I don't know how far you are familiar about drug delivery, let me answer from basic.
1. Prepare standard drug solution of different concentrations. (Ex. 0.2, 0.4, 0.6.... 2.0 mg) in the same buffer that you have used in the bath.
2. See the absorbance of each drug solution and plot the graph. You will get a linear equation.
3. Now, from the 150 ml bath (where 5ml dialysis bag along with conjugated drug was placed), pipette out 2 ml of solution and check for absorbance at an interval of every 30 minutes and fill the bath with blank buffer (Disputable concept though).
4. Plot the values in graph or substitute the absorption value in linear equation. You will get the concentration of drug in the bath.
5. From the concentration (x/150ml), you can calculate the amount of drug released.

Thank You all. Himanshu Bhatt i want to know what is 5 x and 900 x here and why u have taken 5x* D from the second reading. and how much sample have been withdrawn ? is it 5 ml ?
5x is dilution factor for the sample withdrawn (1ml) from 900 ml dissolution media
So as i have withdraw 3 ml and 150 ml is the dissolution bath. so i should multiply the value with 3 and 150 respectively . and for calibration curve i have also taken x ug/3ml and then took the absorbance.
Tell me the volume withdrawn, diss media volume, absorbance or area, line equation..i will send you complete dissolution profile..

I have withdraw 3ml after every 15 minutes
diss media volume : 150 ml
equation from standard curve is y=0.0721*x + 0.0044
I have not dilluted the withdrawn sample anymore.
