I'm trying to quantify the total flavonoid content in orange juice using the spectrophotometric method.

So far, I have a good calibration curve using Rutin as a standard, even though, my extracts don't react the same way as rutin. They don't get red after adding AlCl3 sol and NaOH sol., in fact they develop a yellow color after the addition of AlCl3 sol. and NaOH sol.

My extracts are made with 5 ml of orange juice + 10 ml of methanol. They were shook for 1 minute, then centrifuged and I collected the supernatant. I already tried to do all the extraction in the dark using sonificatiin under mild heating (55 ºC), and got the same yellow color.

Does anyone have experience with this analysis? Any ideas?
