Hi,

I have found two SNPs (Alanine to serine & threonine) at different positions in my desired gene (Sanger sequencing), and then I have built a protein 3D (tertiary) structure by using I-Tasser and swiss-model servers. When I replaced SNPs (at mutated positions) with a mutated one, I found an interesting effect on protein structure that it's making a hydrophobic bond there and protein showing the increase in negative delta G (making sense that protein getting more stable) (YASARA tool was used to replace amino-acid and studied the effect of replacing amino-acids). My protein is a glycosylated protein (majorly studied in glycosylated studies) and now, I want to replace or add these desired mutated/SNPs positions with glycosylated amino-acid But I don't know how to replace or add a glycan or phosphate group at desired positions. Kindly, Please tell me the software or procedures to replace an amino-acid with glycosylated/phosphorylated amino-acid.
