Your question concerns the polymers in the solid state. Correct ?
Then most probably - no one of the factors you have listed - will lead to the change in average Mw. Except may be "removing the defects". How will you remove the defects incorporated in the polymer chain ??? - only by eliminating defects in some reaction - then certainly - the Mw will drop down.

With tuning the synthesis process we can control the defects inside the polymeric chain. Sir, I am curious to know, Is the drop in MW of polymer further affect the density ? I think, it should be high. Please explain ?

The Mw will not change if you only influence the chain arrangement or free volume by, for example, melting and re-crystallization or swelling of the polymer. However, if you change the polymerization parameters to obtain different values of the variables that you have listed, then I would think that also the Mw distribution might have changed.
Furthermore, the Mw could also change if you would use high temperatures or some other "harsh" conditions to change the chain arrangement / free volume.
