Acridine orange is an organic compound. It is used as a nucleic acid-selective fluorescent cationic dye useful for cell cycle determination. with an excitation maximum at 502 nm and an emission maximum at 525 nm (green). So what is the protocol of Use acridine orange staining?
First arrest your cells (I.e. formalin or other), unless your doing live imaging, then just leave them in PBS. Make an acridine orange solution of 0.1ug/mL and add it directly to your cells for 20 minutes. You can then directly image, or wash the cells if you think the background is too high (FYI, you can see under any green or orange filter on a fluorescent scope).
