i am currently working on wheat nutrient uptake evaluation on different soil types. but i did not get clear methodology how to extract protein from wheat grain in laboratory. could you help me pls by sharing the clear methodology, i want to practice and do my self the lab work.

I have experience only for plant roots and leaves by phenol extraction method. I am not sure "it would be helpful for you or not".

You should find out the protocols for protein extraction in plants using anf of the following
1. SDS - PAGE method ( Sodium Doedocyl sulphate - Polyacrymide gel electrophresis)
2. You can also try the Kjedahl analysis for protein through Nitrogen extraction method.(this method may obsolete but can still be very helpful)
You can try any of this methods if the facilities needed are available at your disposal.
Dear Dr. Godwin Michael Ubi i used phenol-extration method only for leaf and root protein isolation> then did SDS-PAGE for protein seperation. This method is very handy and eassy.
