When we do dialysis on hyaluronic acid we lose half of the material.
The pore size of the dialysis bag is 12-14 kDA and the Mw of HA is 100-150 kDa.
Why do we lose so much material?
I guess the Mw of HA has a normal distribution and that some fragments will be small enough fo pass through, but that should not account for half of it right?
It has been a long time since I did this in research, but hyaluronic acid can form complexes that stick to the dialysis tubing. Maintaining the dialysis solutions at an acidic pH is necessary, as described in this patent:” according to the present invention, the hyaluronic acid and / or salt thereof including a step of dialysis treatment with an ultrafiltration membrane after adjusting the hyaluronic acid solution containing hyaluronic acid and its salt and impurities to an acidic pH. Salt purification methods are provided. According to this manufacturing method, impurities can be effectively removed.”

Is it possible that material is not passing through dialysis tubing but instead may be adhered to inside of tubing? Perhaps try rinsing and massaging inside tubing with small volume.
