Supposed I need three different concentration for two plant extracts (namely: Aqueous and Ethanolic extract), starting at 50mg/ml, then 100mg/ml then 150mg/ml. In what particular weight of plant powder should I need to obtain such concentrations?
The total volume I need for the whole experiment is 860ml that will be used in Antibacterial test (60ml - accompanied with 2 extracts in 3 bacteria with 10ml each plate), Cytotoxicity test (100ml) and Phytochemical test (500ml - concentration is not needed)

generally, the powder is used at a rate of 10% (m / volume of solvent). but the amount of powder you must use for your experiments depends on the yield. there are more profitable plants than others and it depends on the climate, the harvesting method and the extraction method.

If you want to study the biological activities (in vitro) and determine the chemical profile (by LC/MS) of your crude extracts (Aqueous or Ethanolic extract) I think 100g of plant powder (from leaves or flowers or roots) for each solvant extract, is enough. But if you want to isolate pure compounds and study pharmacological properties (in vivo) you will need big quantity of plant powder.

It will depend on the yield of each extract from the plant powder. You must have enough of each extract before starting your experiments. Generally in vitro studies do not require a large amount of extract as opposed to in vivo studies where concentratios are expressed as a function of body weight. In addition the consumption will be as great as the experimentation is long.
