I want to use proteomics on mice sclera and western blotting to confirm the proteomics results . However, I am not sure how much mice sclera I need to get enough protein to be able to perform proteomics analysis and to perform western blotting. I aimed for an N=3 per genotype, so I will have 3 mice eyes to perform proteomics and also 3 eyes for western blotting. Hereby seperately lysing the sclera of each eye and performing proteomics seperately on each sample. I know this amount has been used for guinea pig sclera, but as the mice eye is approxiamety 3x as small, I am not sure if I will get enough protein. Does any one have any experience with using mice sclera for such methods? Any tips are welcome.
