Not at all recommended to use the samples stored at 4 degree Celcius. And even though if you are talking about -4 degree Celcius, it not good. At least the samples should be stored at -20 degree Celcius or the best is -80 degree Celcius.

Assuming you are planning to look at mRNA, then you need fresh cells. Most messenger RNA in E. coli will turn over quite quickly.
