I am currently culturing human astrocytes in full serum but I read it is recommended to starve the cells before treating them. Could anyone tell me how long these cells would last in Astrocyte media +1% FBS? Thank you :D
I have experience in rodent astrocytes. Astrocytes resist very well starvation. Before every treatment I performed a short starvation of 30 min - 1 hour (medium without FBS) so the FBS doesn't interfere with the treatments. I didn't tested longer starvation periods with primary astrocytes, but astrocyte cell lines resist 24 hours of starvation without problem (no autophagy detected).
I don't recommend to use FBS (1%) along with the treatment, the effect of your treatment could be shadowed by the FBS.
I hope someone with experience in human astrocytes could give you some other tips.
Thank You Raul, I tried 6 hours and noticed no visible signs of stress so I will try 24 hours and see. My other cells don't seem troubled with 24 hours of starvation, so I will try it with the astrocytes. I will also try removing the FBS, I agree with you, it may mask the effects of my treatment.
