The samples of fish skin were collected from a constant area in the left side of the fish between the ventral and dorsal regions and the fat layer was then removed from the skin samples. Skin samples were weighed and the carotenoid was extracted using acetone, until no colour was observed anymore. Partial evaporation of the combined acetone extracts was performed until 5 mL was approximately attained. Five millilitre of n-hexane and 2 mL of water were added and the mixture was shaken and allowed to separate. Then, the hypophase was washed by n-hexane until no colour was observed. After pooling, the organic epiphases evaporated and were dried by a stream of nitrogen. The extract was dissolved in a known amount of n-hexane. To measure carotenoid in n-hexane, the absorbance was read at 470 nm, and then the concentration of carotenoid is estimated.

First of all I want to thank you all for your reply.

Ornamental fish business depends upon colour.

Conditions:

1. In my experiment suppose I am not allowed to kill the fish.

2. I am going to compare fish in the 1000 lx (unit of illuminance). For example the lux (symbol: lx) is the SI derived unit of illuminance and luminous emittance. lx meter is available in market.

3. Phenotypically all fish is same size, shape and color i.e. red.

4. Now which one is looks good I want to grade for example: price tags 100 $, 50 $, & 10 $.
