I want to study the Seed Germination of Desert Plants under Different Water Potentials due to Drought and salt. i am using 54, 109 and 164 mM NaCl, how i can measure osmotic Potential of these solution and Prepare PEG solution of Equivalent osmotic potential.

In order to measure osmotic Potentials of the three NaCl solutions, you have to use osmometer. To prepare the osmotic potentials equivalent to that of NaCl solutions using PEG, add PEG to your medium and measure using the osmometer until you get the needed solute potential.

PEG doesn"t go in the cell. NaCl the ions with time go in changing the stress. You may have o look at: Astarita ; Ferreira & Bergonci. Allelopathy J. 3(1): 43-50. 1996. The article mentioned by dr. Jose Carneiro was written down in portuguese, is very good, but you may have some difficults in follow it..
