I have used 2.5 g plant tissue for extraction and then further proceed for S.P.E 1 ml extract. Similarly, I have tested my water sample and their unit is µg/L. I am a bit confused about my estrogen concentration unit in plant tissue. I am wondering how I can calculate accurate unit of my plant sample.

Is the molecule actually 'estrogen' (USP has a primary standard) or estrogen-like?
You will have to 'defend' your position (GC/MS is known to misidentify molecules). The answer is;
ug/L = ng/mL (since there are 1000 ng in a ug, and 1000 mL in a L), Therefore,
ng/mL x 1 mL (used in SPE) = ng
ng x mL of aliquot (which you did not specify) = total number of ng in aliquot
[total number of ng in aliquot x (mL extraction fluid/mL aliquot)]/2.5 g = ng/g (or ppb
"Estrogen" is a generic term and is not a specific compound. Mammalian estrogen includes estradiol17B, estradiol17A, estrone and several other compounds, including the sulfate forms of these compounds in urine and feces. One would not expect to find these pure compounds in plants, but there are many, many different estrogenic compounds in plants, fungi and other organisms. If you say the molecule is "actual estrogen", you have not defined it, because there is no specific compound that identified as estrogen.
