I'm trying to grow rice because it has only one copy of my GOI as opposed to Arabidopsis. It plants do germinate (at least WT) and I got some seeds but it took more than 6 months and again, from my mutants I didn't get almost any panicles and even the WT plants had almost empty seeds (not fertilized probably). Here are my questions:
It seems that there is not much difference in germination rate between placing the seeds directly into soil and surface-sterilize and keep them on filter paper for two weeks. However, with the sterilization I can directly see how many germinated, but the plants seemed somewhat smaller in the beginning. So which is your preferred way?
In some manual they wrote they water the plants like twice a week with nutrients and once per two weeks they use nutrients with iron. Is the iron really so important?
Thing that got me stuck probably most - water level. In the manual they wrote the pots should not stand in water for first two weeks or so, it should be just watered extensively. IS that OK? When should I keep it in water? When should I keep the water level above soil? Or should I at all? We have the symbiotic plant so it should be OK: And when should I leave the water out? When the plants start flowering, earlier, later? Because right now I'm waiting already for third harvest from my plants, but I don't know whether it wouldn't help to fill the seeds or something if I've let the plants dry.
Flowering and giving seeds is another chapter. Is there any way to promote (accelerate) flowering in the greenhouse? I just checked my plants in the greenhouse and they are starting to flower and we have light for 12 hours here. In the growthchamber it's easier to control the daylength, but the problem is, that the plants are very tall and they're reaching the lights and burning itself. The plants in the growthchambers have about 1.5 meters, while those in the greenhouse have only ~0.5 meter, which one is more correct?
And is there any way to help the plants to give seeds? Because especially with the mutants I got like 1 seed per plant, that's not very productive...
Hi, I think we can help you more efficiently if you could provide what cultivar or species of rice you're working with so we could give you specific handling. :) But usual is that you can sow it directly n the soil or pre-germinate it for 3-5 days then sow it. You need to transplant it 21 days after sowing. you can maintain water level at 5cm till maturity. Basal application of Fertilizer is 10-14 days after transplanting and final application is at 56 days after sowing with complete fertilizer (NPK) following the recommended rate, you can adjust the amount depending on your soil fertility level.. Also for the growth and development of rice, you need to consider or take note of what cultivar or rice species you are using and where or what environment you are growing it with.
As for your mutant, sterility is one effect of mutation. What kind of mutagen have you used? I can send you files for rice production if you want...Hope it helps... goodluck in your studies

so they should be Nipponbare, ecotypes Hwanyoung (and Dongjin, but I'm not interested in that one anymore).
By keeping the water level at 5 cm you mean under the pot or above the soil?
They are T-DNA insertion mutants, not produced by me. Of course I'm aware (and scared:) that it can be a phenotype.
BTW shall I keep the seeds in seed coat during storage or is it OK to get them out to see how many exactly I have?

Its above soil level. I had experienced growing Nipponbare under screenhouse/glasshouse condition (Philippine condition) and its really short about 35 to 50 cm.
You can always use your wildtype as your reference weather or not the sterility is due to your mutant's phenotype or just environmental effect :)
seed coat meaning, the seed package material or the rice hull, if its rice hull, keep the seed coat, dehulling your seeds for storage it might affect your seeds viability/shelf life.

Yes, you need to dry the seeds. Moisture content should be at least 10% or 6% if to be stored long term.
Yes, we don't apply iron. It depends on your soil condition or as required by your material.
Rice is self pollinated. We only do pollination if we're doing hybridization/generation of F1 seeds.
OK, I'll give it probably one more chance, but our greenhouse is pretty bad and cold during winter because it's for other crops and cannot be regulated separately (new, state-of-the-art)
Which season you want to grow rice is very important. if it is in dry season (Boro season) we need to soak it for 24 to 48 hours. It may vary variety to variety. When soak seeds start sprouting keep it up from water then dry out water and keep it in warm condition to germinate. you have to check water level and watering if need. Complete dryness may damage embryo. Proper sprouted seed is subject to disperse in the main seed bed. Disperse seeding is required. Seed amount 25-30g is better for 1 m2 area. At the age of 30 days seedlings it is appropriate to transplant in the main field. In this way you can get vigorous seedling. Proper balance fertilizer, irrigation and management practices will give you good harvest

Rice is the same wheat growing but it need water and hot conditions.
