I am trying to conjugate a peptide molecule of size ranging from 10 -40 KDa to the silver nanoparticles size of 40-120 nm (from SEM analysis). For this I am using charge dependent binding of the protein (at pH 8.0) to the nanoparticles, still I am unable to get a prolonged stability of the conjugates (Silver Nanoparticle-peptide), to maintain stability would you please suggest any protocol for the nanoparticle protein conjugation.

It is a bit hard to answer without knowing how do you prepare your synthesis your NP.

After amine/carboxyl/thiol modification of the NP, you can use crosslinking or even try to conjugate it by creating acyl-chloride of modified NP with carboxylic acid: http://www.masterorganicchemistry.com/2011/12/03/reagent-friday-thionyl-chloride-socl2/ (never tried it before since crosslinking works well enough)

If you have prepared the Ag nano-particles in chemical / bio-chemical method, after completion of synthesis process, before separating Ag nano-particles, mix the peptide, in the same liquid. Proteins / peptides have strong affinity towards metals. Thanks Omri Haifler , I am synthesized the nanoparticles by both chemical (sodium borohydrite and Sodium tricitrate solution) and Biological method by using plant extract. The article is so useful that provided by you, i will work out on that.
I agree with Omri that you have to first functionalize the metal surface with amine/carboxyl/thiol groups and then use click chemistry to attach the peptide.
other strategy could be to generate strong complexes using linkers with highly specific biomolecular recognition like streptavidin-biotin, antibody-antigen, carbohydrate-selectin, etc.

Dear Omri Heifler Juan C. Fraire Vyshnava Satyanarayana Swamy , Could you help me out with any protocol about how to modify NP surface with COOH at the first place. I will use EDC/ NHS crosslinker to conjugate antibody with my nanoparticle BUT I am stuck at finding any protocol to surface modify my NP with COOH in order to use EDC/ NHS.
BTW, I have silver nanoparticle.
