Recently I read an article (Optimization of protein precipitation based upon effectiveness of protein removal and ionization effect in liquid chromatography-tandem mass spectrometry- Journal of chromatography B,785 (2003) 263-275) where it compares the effectiveness of different protein precipitating methods. It concluded that (1:1) aqueous zinc sulphate heptahydrate (10%, w/v): 0.5N sodium hydroxide is the most efficient method for this purpose. I am trying to replicate this precipitation reagent but when I try to prepare it, specifically when I mix the zinc solution with the NaOH solution a precipitation appears. I think Zn is incompatible with alkaline medium.

Can anyone explain to me how can I prepare this protein precipitation reaction please? It would be very important for me to replicate this formula.
