I want to test the food colorant E171(titanium dioxide) on some human lung fibroblast for my bachelors thesis. I have bought some TiO2 99+% powder but I wish to know how to make the solution and dosage. Any hints and insight on this matter is welcome!
I would suggest a saline suspension of 2.5mg/ml - 5mg/ml.
you should sterilise the suspension by autoclave and vortex before application.
TiO2 is very insoluble.
TiO2 is soluble in concentrated HF and H2SO4 so this is not suitable for biological studies.
2.5-5 mg/ml can be too much. We usually use 1 mg/ml.
You can albo buy nano-TiO2 suspension in water i.e. from NanoAmor. We used that also for microbiological tests. 0.75% solution is still active and nearly transparent. http://www.nanoamor.com/metal___oxide_nanopowder_dispersions
I need a solution of 1 mg / ml concentration, and then via dilutions to get 25 , 50 , 75 and 100 ul/ml final concentration because those are the dosages that I want to test onto the cells. A paper that I found recently stated that they made the solution in the growth media and/or PBS , is that possible ? I am using a HEM F12 media supplemented with 10% bovine serum. Also, the cell culture I am working on is HFL 1 , human fibroblasts.

PBS = Phosphate buffered saline (pH and concentration depending)
HEM F12 (perhaps corectly should have been: HAM's F12 medium, a (content of that nutrient fluid variable from suplier to supplier)

Yes, I think it is possible, provided that you are not willing to obtain a real solution, but a suspension of TiO2 particles in your culture media.
