I'm new to the field, in a new lab, and looking for macrophage lines that would be best to use for some in vitro work involving adding tumor cells grown/ modified in vitro to macrophages. There are so many possibilities. Or, is it good to use primary macrophages? How do you pinpoint what lines (or primary cells) are best for cancer work? In my particular cancer field, it seems like most work is in vivo, so just perusing those papers hasn't gotten me very far. Thanks for any tips.
Most if not all of the cell lines that are used are leukemias or lymphomas. As such they are suboptimal models for primary myeloid cells. It is relatively easy to obtain or differentiate monocytes, macrophages, PMNs and DCs from blood or marrow. My suggestion is that they make better and more rigorous models.
I think that THP-1 which is a human leukemia monocytic cell line was used for studying monocyte/macrophage functions and this is act as common model to estimate modulation of monocyte and macrophage activities.
