Hi, in culture of 4-5 DIV you can see the outgrowth of the axon over dendrites. Dendrite are thicker as they are close to the cell body. However, the best way is Tau immunostaining (Axons) and MAP2 for dendrites. Also in mature neurons over 12 DIV (depends of the density of the culture) the presynaptic markers Piccolo and Bassoon localized in a punctate pattern along axons. All the best

Couldn't agree less wit Viviana. MAP2 for dendrites and Tau for axons are the right markers. You should be careful in NOT using Phosphorylated-Tau antibody as it also marks the dendrites. Good luck :)
I'll join the chorus and agree: MAP2 and Tau are right on, and emphasize Narayan's comment that you should make sure not to use a phosphotau antibody. Cheers!
axons are usually thinner, elongated and necessarily have smooth surface throughout the axon. Dendrites (in maturer cultures) are shorter and have a rough surface due to spines or (if not spiny) due to other contacts.
One more question - can anyone recommend good MAP2 and tau antibodies that would recognise human? Thanks again.
map2 works really nicely to stain dendrites. to recognize axons i would prefere SMI-312. it's more specific than tau. otherwise you can use Ab that recognize the AIS (Axon inital segment) such as Ankyrin, spectrin IV

Can anyone recommend a good antibody against non-phosphorylated Tau for axonal marker only ? Thanks a lot.
As a total tau polyclonal antibody, I would recommend the Dako A0024. It gives really nice signals in both WB (http://www.ncbi.nlm.nih.gov/pubmed/24788298) and IF (http://www.ncbi.nlm.nih.gov/pubmed/22961084):

Is there anybody to have experience with staining of rat primary sympathetic cells?
I need some specific dye for axon through sympathetic nerves. I used TH, but it stains the dendrite also.
